Sensors, comparative pressure measurement and endpoint methods that make a freeze-drying cycle measurable and repeatable.

A repeatable freeze-drying process is one that can be measured, not just one that finishes on a timer. Process analytical technology (PAT) turns the dryer from a black box into a source of evidence about what is happening to the product in real time.

Product temperature is the critical variable

Product temperature, not shelf temperature, determines whether the product stays below its collapse limit. Thermocouples give a direct reading but only for the vials or trays they touch and can slightly alter the freezing behaviour of those samples. Wireless and comparative methods complement contact sensors.

Comparative pressure measurement

A Pirani gauge and a capacitance manometer read pressure differently in the presence of water vapour. Comparing the two signals during primary drying gives a real-time indication of whether sublimation is still active — a widely used, non-invasive way to watch the endpoint approach.

Endpoint determination

The end of primary drying is not the moment the shelf temperature is raised; it is the point where the ice has been removed to the required level. Endpoint methods include product-temperature rise, pressure-rise testing and comparative-pressure behaviour. Secondary drying then continues the removal of bound moisture to the target residual.

Regulated use. For sterile or validated processes, these sensors and the recipe that uses them are part of the data-integrity and qualification discussion, defined by the intended use and the required records.
